Click only once on a specific pixel range

Hi Guys,

 

I'd like to know if it is possible control on a sprite with 300x300 pixel to be click only once.

 

for example.

my sprite is 300x300 pixel and I'd like the pixel from 0x100 to be click only once and give a warning

that it has been already clicked.
